The opener has accepted the role after Dane Vilas decided to step down after leading the county for the last four seasons.
Jennings, who has also signed a one-year contract extension with the Red Rose, becomes the county's 37th permanent men's captain.
"I am so proud and honoured to have been offered the opportunity to become the next captain of Lancashire a club which I have made my home since arriving at Emirates Old Trafford in 2018," said Jennings, who was the club's leading runscorer in last season's LV= County Championship, earning him a recall to the England Test squad over the winter.
"Ever since joining, I have been fully aware of how proud this club is of its history and the passion which our members, supporters, players, and staff all share for the Red Rose.
"I am extremely proud to be leading this club into a new chapter and I hope we are able create more history together."
Jennings, 30, has made 131 appearances for Lancashire in all three formats since joining from Durham five years ago.
